Answer
Nope. Amazon prime is like a membership. You can opt for it.
For the 1st month, you would get a free trial of Prime membership to enjoy the features it offers. Then later after one month you decide whether to keep it or not. Basically prime membership consists of few stuff I.e:
- Free 2-Day Shipping with no minimum order size
- Unlimited instant streaming of thousands of movies and TV shows with Prime Instant Video
- 1 book each month from the Kindle Owners' Lending Library
Without Amazon prime you can still all other stuff.
Almost same procedure with the books transferring. Just hit the send to device button and send it to Kindle Fire for reading purpose.
I would suggest you to read these comments from an actual user of Kindle Fire, mentioned about prime membership also:

Hope this helped!

Can someone explain exactly what Amazon Prime includes?
Jacrose
Does it include watching TV shows and movies at no additional costs? Like Hulu Plus sort of? And what about if you get a Kindle...i heard you could get thousands of free books. Is that true?
Answer
Here is the explaination from Amazon:
Amazon Prime is a membership program that gives you and your family unlimited fast shipping, such as FREE Two-Day shipping to street addresses in the contiguous U.S., on all eligible purchases for an annual membership fee of $79. Amazon Prime members in the U.S. can enjoy instant videos: unlimited, commercial-free, instant streaming of thousands of movies and TV shows through Amazon Instant Video at no additional cost. Members who own Kindle devices can also choose from thousands of books -- including more than 100 current and former New York Times Bestsellers -- to borrow and read for free.
So you can watch TV shows and movies at no additional costs and borrow books from the Kindle Owners' Lending Library free with no due dates.
